Noun + をください asks for something: 水をください, "water, please"; これをください, "I'll take this one". It's the everyday way to order in a café or buy something in a shop.
A number goes after the thing, just before ください: りんごを三つください, "three apples, please".
It's polite but direct. お願いします is softer and very common for orders: コーヒーをお願いします. Among friends, ちょうだい. In a shop, pointing and saying これをください is a perfectly good first sentence.
The same ください after a verb's て-form means "please do", a separate point later in the deck.
Register: Polite but direct. お願いします is softer; ちょうだい is casual.
